Abstract

Peripheral artery disease currently affects over 100 million people globally. Balloon angioplasty is commonly used to reopen arteries presenting with severe atherosclerotic occlusion; however, this causes mechanical stress upon the vascular wall and restenosis of the vessel following intervention. Drug coated balloons (DCBs) offer an option for delivery of therapeutics to prevent restenosis, but current DCBs have limited efficacy and off target cytotoxic effects. We present progress towards the establishment of a Niban mimetic (NiPp) peptide DCB platform which delivers within vasculature for the goal of preventing restenosis of arteries following angioplasty. Utilizing a layer-by-layer approach, coating conditions were optimized for NiPp loading and release from the balloon surface. The resulting NiPp-DCBs were delivered in ex vivo vascular tissue and evaluated for their ability to transfer NiPp into vasculature as well as retain peptide within vessels over an extended period.
